Abstract:

Our goal is to develop enhancer virus tools that can provide access to select cell classes, subclasses, and types brain- wide. Enhancer-adeno-associated viruses (enhancer AAVs) will be screened for labeling specificity by whole-brain light sheet imaging. Datasets are produced by axially-swept light sheet microscopy of whole mouse brains injected with enhancer AAVs using the SmartSPIM (LifeCanvas) platform. Cell detection and anatomical placement are performed on the full-resolution three- dimensional images. In addition, we generate a series of maximum intensity projections (MIPs), representing 30 µm of depth at 100 µm spacing from the full-resolution three-dimensional images, that can be quickly and consistently visually evaluated for overall expression strength and distribution.
